i dont know after effects - but this may help

How much free space do you have on the PC?
with photoshop - you can set how much memory you can use in preferences - can you do this with after effects
how much scratch space is assigned for after effects
How big is the file you are trying to open

Okay, a couple of things---First is you dont want AE sitting on the same drive that houses the video files you want to play with.

Edit/File Preferences/Cache/

Set it to 60% Set Maximum to 120%
