You will need to create an algorithm to create and check the serial numbers, then when the program runs for the first time use the algorithm to verify that the program user has a valid serial number.

To expand a bit on what was already said you can also use Visual Basic code to create a new registry key value to indicate whether or not the software has been registered. When the program runs each time it would look to see if the registry value has been created and prompt the user for code as needed.

It would also be worthwhile, if you go down the registry line, to make the key obscure enough that you can't just do a search for it and find it to set it to true on another computer. Mind you this would only be necessary if you are tracking the serial keys online or somewhere else they can be verified as not being used.
