Error Message at startup (WINXP)

At startup I keep getting this message headed "16 bit windows subsystem".
E:\WINDOWS\SYSTEM32\AUTOEXEC.NT. The system file is not suitable for running MS-DOS and Microsoft Windows applications. Choose 'Close' to terminate the application.
What does the message mean, and how can I stop it happening every startup? :confused:
